Indications/Uses
Symptomatic relief of nausea & vomiting of functional, organic, infectious or dietary origin, induced by radio- or drug therapy & dopamine agonist used in Parkinson's disease.
Dosage/Direction for Use
Adult & adolescent ≥12 yr, ≥35 kg & childn <12 yr, ≥35 kg 10 mL tds-qds. Max: 40 mg daily. Adult & adolescent ≥12 yr, <35 kg 0.25 mg/kg tds-qds. Max: 35 mg daily.
Administration
Should be taken on an empty stomach: Take 15-30 min before meals.
Contraindications
Hypersensitivity. Prolactinoma. Patients w/ known existing prolongation of cardiac conduction intervals, significant electrolyte disturbances or underlying cardiac diseases eg, CHF; presence of GI haemorrhage, mechanical obstruction or perforation. Co-administration w/ QT-prolonging drugs, potent CYP3A4 inhibitors. Moderate or severe hepatic impairment.
Special Precautions
Discontinue use if signs & symptoms of cardiac arrhythmia occurs. Increased risk of serious ventricular arrhythmias or sudden cardiac death; proarrhythmia. Not to be taken by patients w/ lactose/sorbitol intolerance, galactosemia or glucose/galactose malabsorption. Not to be taken simultaneously w/ antacids or antisecretory agents. Concomitant use w/ QT-prolonging drugs, potent CYP3A4 inhibitors, antacids or antisecretory agents. May affect ability to drive & use machines. Severe renal impairment. Pregnancy. Not recommended during lactation. Infant & childn <12 yr, <35 kg.
Adverse Reactions
Depression, anxiety, decreased/loss of libido; headache, somnolence, akathisia; diarrhoea; rash, pruritus; breast enlargement, tenderness & pain, galactorrhoea, amenorrhoea, irregular menstruation, lactation disorder; asthenia.
Drug Interactions
Increased plasma levels w/ CYP3A4 inhibitors. Antagonized anti-dyspeptic effect w/ anticholinergic drugs eg, dextromethorphan, diphenhydramine.
